In Vitro Didanosine is converted by cellular enzymes to the active antiviral metabolite dideoxyadenosine triphosphate (dd-ATP) and the intracellular half life of dd-ATP varies from 8-24 hours.
Didanosine shows antiretroviral activity with IC50 value of 0.24-0.6 mg/L for HIV infection.
Didanosine (5, 10, 50, 100 ug/ml; 24, 48 h) shows no significant inhibition of cell proliferation in IEC-6 cells.
In Vivo Didanosine (100, 150 mg/kg; p.o.; daily for 7 days) decreases the length of duodenal and in jejunal villus in mouse.
