Description | 2-deoxy-D-glucose is a non-metabolizable glucose analog that inhibits phosphorylation of glucose by hexokinase, the first step of glycolysis. This results in the depletion in cellular ATP, the inhibition of protein glycosylation, and the disruption of ER quality control by inducing the unfolded protein response. 2-deoxy-D-glucose has been shown to cause cell cycle inhibition and cell death in in vitro models of hypoxia, induce autophagy, increase reactive oxygen species production, activate AMPK, and block tumor cell growth in animal models. |

In Vitro | 2-Deoxy-D-glucose significantly reduces the level of ATP in MCF-7 cells that paralleles the effects of 2-DG on cell growth. The levels of phosphorylated Akt are significantly decreased, whereas the levels of phosphorylated AMPK and Sirt-1 are significantly increased in MCF-7 cells exposed to 2-Deoxy-D-glucose. 2-DG treatment increases the levels of pentose phosphate pathway (PPP) metabolites and augments the generation of NADPH by glucose-6-phosphate dehydrogenase. |
In Vivo | 2-Deoxy-D-glucose causes a 7% decrease in final weight, and delayes the appearance of palpable mammary carcinomas. 2-Deoxy-D-glucose is decreased in a dose-dependent manner in rat muscle. |
